responsive nanocomposite hydrogel in sensitized osteosarcoma chemotherapy and immunotherapy

Sequential responsive nanocomposite hydrogels are biomaterals with photothermal effects and controlled release functions that can be used in chemo therapeutic and immunotherapy for sensitized osteosarcoma. Nanocomposite injectable hydrogel system based on the near infrared light response for the delivery of immune checkpoint inhibitor molecules: The near infrared light-responsive nanocomposite injectable hydrogel system can be used to deliver PD-L1 antibodies and achieve their long-term controlled release. This system showed a good inhibitory effect on the postoperative anti-recurrence model of osteosarcoma while promoting bone regeneration. This system has good biocompatibility and safety, providing a new strategy for synergistic treatment of osteosarcoma. Nanocomposite multifunctional hydrogel based on magnesium oxide nanoparticles and 2-aminoethyl grafted polydopamine shell: Bioactive nanoparticles (MDA-NPs) composed of magnesium oxide nanoparticles (M-NPs) core and 2-amino (2-AM) grafted polydopamine (PDAM) shell, prepared with phosphonate ester modified methacrylamide chitosan (CMP) and polyacrylamide (PAM) into a series of MDA-NPs nanocomposite materials. This hydrogel shows equilibrium photothermal effects, mechanical properties and osteogenesis, and MDA-NPs can be used as co-crosslinkers, photothermal agents and Mg2 + reservoirs. The hydrogel and near-infrared (NIR) laser irradiation can effectively inhibit human osteosarcoma cells (143B), achieving complete inhibition of tumor recurrence in vitro and in vivo. The released Mg2 + effectively promoted the osteogenic activity of mouse embryonic osteoblast precursor cells (MC3T3-E1), and the hydrogel showed highly efficient bone repair performance in a rat model of severe cranial defect. Nanocomposite hydrogel prepared based on metal-organic frame (MOF) and polyethylene glycol dimethyl acrylate (PEGDMA): nanocomposite hydrogel prepared from metal-organic frame (MOF) and polyethylene glycol dimethyl acrylate (PEGDMA) can be used to deliver multijobicin (DOX) and platinum drug cisplatin (CDDP) to achieve synergistic chemotherapy for osteosarcoma. This hydrogel is well biocompatible and injectable, and can form a gel at the tumor site and release the drug under the excitation of NIR light. This hydrogel showed an efficient killing effect of osteosarcoma cells both in vitro and in vivo, while facilitating the repair of bone defects.
